DESCRIPTION:

Clindamycin is a lincosamide antibiotic, which is a chlorinated derivativeof lincomycin with a primarily bacteriostatic action against Gram-positive organisms and a wide range of anaerobic pathogens, as well as some antiprotozoal actions. It used, usually where other drugs are unsuitable, in the treatment of anaerobic, staphylococcal and streptococcoal infections, and in the prophylaxis of endocarditis. It has also been tried in Pneumocystis carinii pneumonia and in protozoal infections such as babesiosis, malaria and taxoplasmosis. It is given by mouth as the hydrochloride or palmitate hydrochloride, or parenterally as the phosphate: It is also applied topically in the treatment of bacterial vaginosis and of acne.

FORMULATION:

Each capsule contains:
Clindamycin (as Hydrochloride ………………. 300 mg)

PHARMACOKINETICS:

About 90% of a dose of Clindamycin hydrochloride is absorbed from the gastrointestinal tract: concentrations of 2 to 3 mcg per mL occur within 1 hour after a 150 mg dose of Clindamycin, with average concentrations of about 0.7 mcg per mL after 6 hours. After does of 300 and 600 mg peak plasma concentrations of 4 to 80 mcg per mL., respectively. Have been reported. Absorption is not significantly diminished by food in the stomach but the rate of absorption may be reduced. Clindamycin palmitate hydrochloride is rapidly hydrolyzed following oral administration to provide free Clindamycin.

ADVERSE EFFECT:

Adverse effect include diarrhea in some patients antibiotic-associated colitis has occured, and may prove total. Other adverse effects include skin rashes and other hypersensitivity reactions, blood dyscrasias and disturbances of hepatic function.

DRUG INTERACTION:

The use of antidiarrheal agents which reduce peristalsis, such as diphenoxylate, loperamide or opioids and in patients with antibiotic associated colitis may exacebate the syndrome by delaying excretion of the toxic. For the effect of kaolin-pectin suspension on Clindamycin it was found that the suspension had no effect on the extent of Clindamycin absorption but did markedly reduced he absorption rate.

INDICATIONS:

It is indicated in the treatment of anaerobic, staphylococcal and streptococcal infections, and in the prophylaxis of endocarditis. It has also been tried in pneumocystis carinii pneumonia and in protozoal infection such as babesiosis, malaria and toxaplasmosis.

PRECAUTIONS:

Clindamycin should not be given to patients hypersensitive to it or to the closely-related drug lincomycin. It should be used with caution in patients with gastrointestinal disease, particularly those with history of colitis. Clindamycin should be withdrawn immediately if significant diarrhea or colitis occurs. Caution has also been adviced in atopic patients, in those with hepatic or severe renal impairment who may require dosage adjustment. Clindamycin is also excreted is breast milk and breast-feeding should be avoided during therapy.

DOSAGE & ADMINISTRATION:

Mild to moderate infections:
1 Capsule every 6 hours or as prescribed by the physician.

CAUTION:

Food, Drugs, Devices and Cosmetics Act prohibits dispensing without prescription.

STORAGE CONDITION:

Store at temperatures not exceeding 30 degree Celsius.
